At a distance of 2 cm directly above one end of the plate, a small spot is applied for the solution of the mixture to be separated. A plate is later placed in a closed jar that contains a fluid known as eluant. Doing this, the plate then rises up carrying several components of the mixture to different heights.
Column Chromatography
This technique is used to separate the components of a mixture by using a column of suitable adsorbent filled in a glass tube. An appropriate eluant is slowly made to flow by placing the mixture on the top of the column.
The separation of the components takes place according to the degree of adsorption of the components present on the wall adsorbent column. Thus, the highest absorptivity component is retained on the top, whereas the other components flow down to different heights.
Partition Chromatography
In this technique, a continuous differential separating of components of a mixture takes place into a mobile phase and stationary phase. The example of separation chromatography can be seen in paper chromatography technique. In this process, the chromatography paper is used as a stationary phase that acts as a mobile phase, owing to the suspension of solvent in a mixture.
When a spot is placed at the base of the chromatographic paper and as the solvent rises up the paper, the components are passed to different degrees depending upon their holding. Thus, the components are separated at various heights.
